Nanoparticle Characterization

Nanoparticle characterization is the systematic measurement of a nanoparticle’s physical, chemical, and biological properties, which determines how it behaves in research and clinical environments. Researchers assess features such as particle size, morphology, surface charge, composition, aggregation state, and interactions with surrounding media using complementary methods including electron microscopy, dynamic light scattering, spectroscopy, and zeta-potential analysis. In bioengineering, these measurements guide the design and quality control of nanoparticles for drug delivery, imaging, biosensing, tissue engineering, and other biomedical applications. Reliable characterization links nanoscale structure to stability, cellular interactions, performance, and safety.

Simple homogenization was used to prepare novel, high-density, lipoprotein-mimicking nanoparticles to encapsulate nerve growth factor. Challenges, detailed protocols for nanoparticle preparation, in vitro characterization, and in vivo studies are described in this article.

The objective of this protocol is to incorporate SDF-1α, a stem cell homing factor, into dextran sulfate-chitosan nanoparticles. The resultant particles are measured for their size and zeta potential, as well as the content, activity, and in vitro release rate of SDF-1α from the nanoparticles.
