Puncture Injury Induction

Puncture injury induction is a controlled method for creating localized tissue damage with a needle, probe, or similar instrument to model wound formation in biological or engineered systems. The mechanical breach disrupts tissue architecture and can initiate processes such as bleeding control, inflammatory signaling, cellular migration, and matrix remodeling, depending on the tissue and experimental conditions. In bioengineering, researchers use this approach to evaluate wound-healing responses, test biomaterials and tissue-engineered constructs, and assess how engineered tissues recover after injury. Standardized puncture size, depth, and location improve experimental reproducibility and enable comparisons among treatments or model designs.

Source: Macheda, T., et. al. Electromagnetic Controlled Closed-Head Model of Mild Traumatic Brain Injury in Mice. J. Vis. Exp. (2022)This video demonstrates the procedure to induce a closed-head injury in a mouse model using a stereotaxic frame. The controlled impact replicates mild traumatic brain injury, allowing researchers to study neurological effects and recovery by measuring post-injury reflexes and behavior.

The isolated rat heart is an enduring model for ischemia reperfusion injury. Here, we describe the process of harvesting the beating heart from a rat via in situ aortic cannulation, Langendorff perfusion of the heart, simulated ischemia-reperfusion injury, and infarct staining to confirm the extent of ischemic insult.

Induction of Traumatic Brain Injury in a Rat Model Using a Lateral Fluid Percussion Device

This video demonstrates the induction of traumatic brain injury (TBI) in a rat using a lateral fluid percussion injury (FPI) device. The procedure involves creating a hole in the skull of an anesthetized rat to expose the dura mater, onto which a cannula is attached. The rat is then connected to the device, which generates a pressure wave that reaches the brain, mimicking TBI by causing neural cell death and vascular...
