In Vitro Thrombolysis

In vitro thrombolysis is the laboratory study of blood-clot dissolution outside the body, providing a controlled way to evaluate how thrombolytic therapies and engineered systems restore vessel patency. In these models, a clot is formed under defined conditions and exposed to fibrinolytic agents, such as tissue plasminogen activator, which converts plasminogen to plasmin; plasmin then degrades fibrin, the structural scaffold of the clot. Bioengineers use in vitro thrombolysis assays and flow platforms to measure clot breakdown, treatment kinetics, and drug or device performance, supporting the design and optimization of safer, more effective strategies for treating thrombosis.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.

In-vitro thrombolysis assays have often struggled to replicate in-vivo conditions whether in the model thrombus being digested or in the environment in which thrombolysis is occurring. Herein, we explore how coupling the Chandler loop and Real-Time Fluorometric Flowing Fibrinolysis Assay (RT-FluFF) is used for high-fidelity, ex-vivo, clot lysis monitoring.

This procedure was established to be used for developing advanced 3D hepatic cultures in vitro, which can provide a more physiologically relevant assessment of the genotoxic hazards associated with nanomaterial exposures over both an acute or long-term, repeated dose regimes.

Whole organ culture of the intervertebral disc (IVD) preserves the native extracellular matrix, cell phenotypes, and cellular-matrix interactions. Here we describe an IVD culture system using mouse lumbar and caudal IVDs in their functional spinal units and several applications utilizing this system.
