Rat Cardiomyocytes

Rat cardiomyocytes are heart muscle cells isolated from rats and used as experimental models for studying cardiac structure, function, and engineered tissues. These contractile cells generate force when electrical excitation triggers calcium entry and intracellular calcium release, linking membrane depolarization to actin–myosin contraction; their rhythmic activity can be monitored in culture. In bioengineering, researchers seed rat cardiomyocytes onto biomaterial scaffolds, engineered substrates, or microfluidic platforms to assess cell alignment, electrical conduction, contractility, and tissue maturation. This model supports the development and testing of cardiac patches, drug-screening systems, and disease-relevant in vitro platforms while showing how material and mechanical cues influence cardiac performance.

Isolation and Cryopreservation of Neonatal Rat Cardiomyocytes

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.

Several protocols have been developed and described for the isolation of different cardiac cell types from a rat heart. Here, an optimized protocol is described that allows the isolation of high-quality major cardiac cell types (cardiomyocytes, endothelial cells, and fibroblasts) from a single preparation, reducing the experimental costs.

Here, we present a protocol for the isolation and cultivation of adult rat ventricular cardiomyocytes (ARVC). Isolated ARVC can be used for short and long-term cultivation. The isolation and cultivation of ARVC can play a key role in developing new treatment regimens for cardiac diseases.

Measuring Fast Calcium Fluxes in Cardiomyocytes

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.
