Hemicellulose Unchanged

Hemicellulose unchanged refers to plant-cell-wall hemicellulose that remains in its native composition and structure rather than undergoing chemical, enzymatic, or thermal modification. Hemicellulose is a heterogeneous group of branched polysaccharides, including xylan and mannan, that surrounds cellulose microfibrils and interacts with lignin through hydrogen bonding and other associations. In bioengineering, preserving this fraction provides a reference state for evaluating biomass processing, pretreatment, and engineered deconstruction strategies. Comparing unchanged hemicellulose with altered material helps researchers assess polymer accessibility, sugar release, mechanical properties, and the efficiency of producing biofuels, biomaterials, and other renewable bioproducts.

Biofuels: Producing Ethanol from Cellulosic Material

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University In this experiment, cellulosic material (such as corn stalks, leaves, grasses, etc.) will be used as a feedstock for the production of ethanol. The cellulosic material is first pretreated (ground and heated), digested with enzymes, and then fermented with yeast. Ethanol production is monitored using an ethanol probe. The experiment can be extended to optimize ethanol production by varying the feedstock used,...

Calcofluor White Staining: A Simple Method for UV-Induced Fluorescence Visualization of Cellulose Deposits in Cell Walls of Dicot Plant Stem Sections

In this video, we describe a staining procedure to visualize cellulose deposition within cell walls of Arabidopsis thaliana, a dicot plant, using calcofluor white, which specifically binds to β-linked glucans, exhibiting fluorescence under UV light.

Conserved Binding Sites

2020

Many proteins’ biological role depends on their interactions with their ligands, small molecules that bind to specific locations on the protein known as ligand-binding sites. Ligand-binding sites are often conserved among homologous proteins as these sites are critical for protein function. Binding sites are often located in large pockets, and if their location on a protein’s surface is unknown, it can be predicted using various approaches. The energetic method computationally analyses the...

2016

A high-throughput assay for enzyme screening is described. This multiplexed ready-to-use assay kit comprises of pre-chosen Chromogenic Polymer Hydrogel (CPH) substrates and complex Insoluble Chromogenic Biomass (ICB) substrates. Target enzymes are polysaccharide degrading endo-enzymes and proteases.
