Receptor Assembly

Receptor assembly is the process by which receptor proteins fold, associate into functional complexes, and become positioned to detect specific signals. It depends on molecular recognition between receptor subunits, supported by noncovalent interactions and coordinated processing or trafficking that establish the receptor’s structure, localization, and signaling competence. In bioengineering, controlling these steps helps researchers design synthetic receptors, optimize cell-based sensors, and study how changes in receptor composition alter signal transmission. Such control can improve the performance and interpretability of engineered cells while clarifying links between molecular architecture and cellular response.

The analytical ultracentrifuge (AUC) sample cell holds sample and reference buffer and during experiments and is exposed to high vacuum and rotor speeds up to 60,000 rpm. This video will demonstrate the rigorous attention to detail necessary for assembly, loading and alignment of this very important component of an AUC experiment.

Protein Complex Assembly

2020

Proteins can form homomeric complexes with another unit of the same protein or heteromeric complexes with different types. Most protein complexes self-assemble spontaneously via ordered pathways, while some proteins need assembly factors that guide their proper assembly. Despite the crowded intracellular environment, proteins usually interact with their correct partners and form functional complexes. Many viruses self-assemble into a fully functional unit using the infected host cell to...

We describe a protocol to observe and analyze cell rolling trajectories on asymmetric receptor-patterned substrates. The resulting data are useful for engineering of receptor-patterned substrates for label-free cell separation and analysis.

High-throughput Analysis of Mammalian Olfactory Receptors: Measurement of Receptor Activation via Luciferase Activity

2014

Olfactory receptor activation patterns encode odor identity, but the lack of published data identifying odorant ligands for mammalian olfactory receptors hinders the development of a comprehensive model of odor coding. This protocol describes a method to facilitate high-throughput identification of olfactory receptor ligands and quantification of receptor activation.
