Tissue Imaging

Tissue imaging is the visualization and measurement of tissue structure, composition, and function, enabling researchers to examine biological organization across cellular to whole-tissue scales. It works by directing or detecting signals such as light, ultrasound, or magnetic responses and converting differences in absorption, scattering, emission, or relaxation into image contrast. In bioengineering, microscopy, optical coherence tomography, and magnetic resonance imaging help evaluate engineered tissues, scaffold architecture, cell distribution, and tissue maturation without relying solely on destructive sampling. These measurements support design optimization, quality control, disease modeling, and regenerative medicine by linking material properties and cellular behavior to tissue performance.

In this manuscript, we present a high-throughput, semi-automated cryohistology platform to produce aligned composite images of multiple response measures from several rounds of fluorescent imaging on frozen sections of mineralized tissues.

The combination of iterative-bleaching-extends-multiplexity (IBEX) and a commercial nucleotide labeling assay (Click-iT EdU) enables the detection and categorization of dividing cell types in highly dynamic processes in fixed frozen murine tissue sections.
