Digital Pcr Assay

A digital PCR assay is a molecular technique that partitions a DNA or complementary DNA sample into thousands of individual reactions, enabling precise, absolute nucleic acid quantification without a calibration curve. After endpoint amplification, fluorescent signals identify partitions containing target molecules, and statistical analysis based on Poisson distribution converts positive and negative counts into starting-copy estimates. Digital PCR supports sensitive detection of rare mutations, copy-number changes, pathogens, and low-abundance transcripts, even when conventional quantitative PCR struggles with small differences or complex samples. Its reproducibility and tolerance of inhibitors make it valuable in diagnostics, environmental testing, and biological research.

In this video, we demonstrate the duplex digital PCR (ddPCR) technique — a modification of the traditional PCR technique that is useful in detecting two different genetic markers simultaneously. A single PCR reaction is partitioned into nanoliter-sized emulsified droplets that are independently amplified, and the detection of differently colored fluorescence amplification signals from the fraction of droplets is used to compute the initial concentration of the target sequences.

We describe an endpoint digital assay for quantifying nucleic acids with a simplified (analog) readout. We measure bulk fluorescence of droplet-based digital assays using a standard qPCR machine rather than specialized instrumentation and confirm our results by microscopy.
