Cell Index Monitoring

Cell Index Monitoring is a label-free technique that tracks changes in cell growth, attachment, and behavior in real time, providing a dynamic view of cellular activity. In impedance-based systems, adherent cells interact with microelectrodes on a culture plate, altering electrical resistance as cell number, spreading, morphology, or barrier properties change. Continuous measurements generate cell index curves that reveal proliferation, cytotoxicity, migration, and other responses without repeatedly disturbing the culture. As a biological technique, cell index monitoring supports drug screening, toxicology, infection studies, and cell-based assays by capturing time-dependent effects that endpoint measurements may miss.

Successful use of cell tracking dyes to monitor immune cell function and proliferation involves several critical steps. We describe methods for: 1) obtaining bright, uniform, reproducible label-ing with membrane dyes; 2) selecting fluorochromes and data acquisition conditions; and 3) choosing a model to quantify cell proliferation based on dye dilution.
