Pyruvate Malate Substrates

Pyruvate malate substrates are paired metabolic fuels used to assess mitochondrial energy metabolism and respiratory function in biological techniques. Pyruvate is converted to acetyl-CoA by pyruvate dehydrogenase, generating NADH, while malate supports tricarboxylic acid cycle activity and supplies intermediates for continued reducing-equivalent production. In isolated mitochondria or permeabilized cells, these substrates drive electron delivery to respiratory Complex I, allowing oxygen consumption to be measured under defined conditions, often with ADP or other metabolic effectors. This approach helps quantify oxidative phosphorylation, identify defects in mitochondrial respiration, and compare how cells or tissues respond to metabolic or pharmacological interventions.
