Fluorogenic Probe Assay

A fluorogenic probe assay is a biological technique that detects a target by converting a nonfluorescent probe into a fluorescent signal, enabling sensitive measurement with minimal background. In a typical assay, target binding or enzymatic activity alters the probe’s structure, such as by cleaving a quencher from a fluorophore, which increases fluorescence in proportion to the reaction or target concentration. Researchers use these assays to measure enzyme activity, detect nucleic acids, monitor cellular processes, and quantify biochemical reactions in real time. Their combination of specificity, quantitative readouts, and compatibility with microplate or imaging platforms supports diagnostics, drug screening, and molecular biology research.

This video demonstrates a flow cytometry assay to quantify the ROS levels in 3D intestinal organoids obtained from GFP-expressing transgenic mice. This method provides an in vitro experimental model to investigate the efficacy of test compounds on ROS production.

A Fluorogenic Peptide Cleavage Assay to Screen the Proteolytic Activity of Proteases

2025

This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission. The fluorescence signal is detected and analyzed to check for the cleavage efficiency of different peptide variants.

2012

We have recently reported a novel approach for generating fluorogenic DNAzyme probes that can be applied to set up a simple, "mix-and-read" fluorescent assay for bacterial detection. These special DNA probes catalyze the cleavage of a chromophore-modified DNA-RNA chimeric substrate in the presence of crude extracellular mixture (CEM) produced by a specific bacterium, thereby translating bacterial detection into fluorescence signal generation. In this report we will describe key experimental...
