Pyruvate Oxidase

Pyruvate oxidase is an enzyme that converts pyruvate into acetyl phosphate, linking central carbon metabolism to acetyl-group transfer and redox chemistry. In the presence of inorganic phosphate and oxygen, the enzyme uses a flavin cofactor to decarboxylate pyruvate, producing acetyl phosphate, carbon dioxide, and hydrogen peroxide. This defined reaction makes pyruvate oxidase useful in biological techniques for coupled enzymatic assays and biosensors that detect pyruvate through measurable peroxide formation. It also supports studies of bacterial metabolism by providing a controlled way to examine pyruvate utilization, oxygen-dependent reactions, and pathways that generate acetyl phosphate.

We describe a modified small-scale extraction and colorimetric assays of lactate and pyruvate in the nematode C. elegans. When utilizing commercial assay kits, the technical development of their sensitivity and accuracy is important. Protein precipitation in extraction is the most critical step for the quantitative determination of intracellular metabolites.
