Actin Conservation

Actin conservation is the evolutionary preservation of actin’s sequence, structure, and cellular functions across diverse organisms, reflecting its central role in biology. Actin proteins bind ATP and assemble reversibly into polarized filaments, while conserved interfaces support interactions with myosin and actin-binding proteins that regulate filament turnover, organization, and force production. This conservation enables cells to maintain essential processes such as cell shape, motility, intracellular transport, cytokinesis, and muscle contraction despite evolutionary divergence. Comparing conserved actin regions helps researchers infer evolutionary relationships, study cytoskeletal mechanisms, interpret mutations, and investigate how actin dysfunction contributes to disease.

Filamentous actin (F-actin) plays an important role in spinogenesis, synaptic plasticity, and synaptic stability. Quantification of F-actin puncta is therefore a useful tool to study the integrity of synaptic structures. This protocol describes the procedures of quantifying F-actin puncta labeled with Phalloidin in low-density primary cortical neuronal cultures.

Conservation of Momentum

Source: Nicholas Timmons, Asantha Cooray, PhD, Department of Physics & Astronomy, School of Physical Sciences, University of California, Irvine, CA The goal of this experiment is to test the concept of the conservation of momentum. By setting up a surface with very little friction, collisions between moving objects can be studied, including their initial and final momenta. The conservation of momentum is one of the most important laws in physics. When something is conserved in physics, the...

Actin Co-Sedimentation Assay; for the Analysis of Protein Binding to F-Actin

Study of the Actin Cytoskeleton in Live Endothelial Cells Expressing GFP-Actin

Microscopic imaging of live endothelial cells expressing GFP-actin allows characterization of dynamic changes in cytoskeletal structures. Unlike techniques that use fixed specimens, this method provides a detailed assessment of temporal changes in the actin cytoskeleton in the same cells before, during, and after various physical, pharmacological, or inflammatory stimuli.

Conserved Binding Sites

2020

Many proteins’ biological role depends on their interactions with their ligands, small molecules that bind to specific locations on the protein known as ligand-binding sites. Ligand-binding sites are often conserved among homologous proteins as these sites are critical for protein function. Binding sites are often located in large pockets, and if their location on a protein’s surface is unknown, it can be predicted using various approaches. The energetic method computationally analyses the...
