Antibody Coating

Antibody coating is a laboratory technique that immobilizes antibodies on a solid surface so they can selectively capture or detect target molecules. During coating, antibodies are attached to materials such as microplate wells through passive adsorption or chemical coupling, while controlled pH, concentration, incubation time, and blocking conditions help preserve binding activity and reduce nonspecific interactions. The coated surface then binds a complementary antigen from a biological sample, enabling signal generation through labeled detection reagents in assays such as ELISA and other immunoassays. By improving target capture and assay specificity, antibody coating supports protein measurement, diagnostic testing, biomarker analysis, and development of research biosensors.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

Overview Antibodies, also known as immunoglobulins (Ig), are essential players of the adaptive immune system. These antigen-binding proteins are produced by B cells and make up 20 percent of the total blood plasma by weight. In mammals, antibodies fall into five different classes, which each elicits a different biological response upon antigen binding. The Y-Shaped Structure of Antibodies Consists of Four Polypeptide Chains Antibodies consist of four polypeptide chains: two identical heavy...
