Automated Counter

An automated counter is an instrument that rapidly measures and records the number of cells, particles, or other biological objects in a sample with limited manual intervention. Depending on the platform, it uses image analysis, optical detection, or electrical impedance to identify objects as they pass through a sensing region, then applies size, shape, or signal thresholds to distinguish them from background and aggregates. In biology, automated counters support cell concentration and viability measurements, culture standardization, microbial analysis, and sample-quality assessment. By improving speed, consistency, and reproducibility, they help researchers monitor experiments and prepare reliable inputs for downstream assays.

The Moxi Z miniature automated cell counter is a novel instrument that combines the Coulter Principle with patented thin-film sensor technology and a proprietary software algorithm to perform sizing and counting of a broad size range of particles as well as to determine the overall health of monodisperse mammalian cell cultures. This protocol describes the use of this instrument for counting and assessing the health of cell cultures.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.

A protocol is described for preparing a simple model ecosystem that recreates the methane-oxygen counter gradient found in the natural habitat of aerobic methane-oxidizing bacteria, enabling the study of their physiology in a spatially resolved context. Modifications to common biochemical assays for use with the agarose-based model ecosystem are also described.
