Bacterial Inoculation

Bacterial inoculation is the controlled introduction of bacteria into a sterile growth environment, enabling researchers to cultivate, examine, or preserve a population of microorganisms. Using aseptic technique, a measured sample is transferred with a sterile instrument into a suitable culture medium, where nutrients, temperature, oxygen availability, and incubation time influence cell division and colony formation. In biology, bacterial inoculation supports isolation and identification of species, evaluation of metabolic and physiological traits, antimicrobial susceptibility testing, and preparation of cultures for molecular or industrial studies. Careful technique helps prevent contamination and produces reliable, reproducible results.

Source: Rufián, J. S., et.al. Single-Cell Analysis of the Expression of Pseudomonas syringae Genes within the Plant Tissue. J. Vis. Exp. (2022)This video demonstrates a vacuum infiltration method to introduce a bacterial pathogen into Arabidopsis plant leaves. The approach uses a prepared bacterial suspension and vacuum pulses to enable uniform entry through stomata, resulting in controlled leaf infection.

Transurethral Catheterization and Bacterial Inoculation to Generate a Urinary Tract Infection Model in Male Mice
