Blue Dye Food Tracking

Blue Dye Food Tracking is a simple biological tracing method that uses a visible blue color mixed with food to follow ingestion and movement through an organism’s digestive system. It helps researchers connect feeding behavior with food passage and gut function. After an organism consumes the labeled food, researchers monitor the dye’s presence and distribution over time, using visible color as an indicator of where the meal has traveled and when it is eliminated. This approach supports studies of feeding, food transit, digestion, and experimental effects on gastrointestinal function, while providing an accessible assay for teaching and research in biology.

Gut Acidification Monitoring Assay: A Technique to Study the Acidification of the Intestinal Lumen in Drosophila using Bromophenol Blue Dye

2025

This video demonstrates an assay to monitor gut acidification in Drosophila flies. After the ingestion of food supplemented with a pH indicator dye, the acidification of the intestinal lumen results in a change in the color of the dye. Visual inspection of the digestive tract of the flies to examine the color change provides evidence of acidification.

This video describes the CApillary FEeder, or CAFE, assay, a behavioral method that measures food intake and preference of unrestrained fruit flies. The featured protocol shows how to perform the assay with minimal evaporation from the liquid-filled capillaries used to deliver the food and track its intake.
