Bundle Of His

The Bundle of His, also called the atrioventricular bundle, is a tract of specialized cardiac muscle cells that carries electrical impulses from the atrioventricular (AV) node into the ventricles, helping coordinate the heartbeat. After a brief delay at the AV node, the impulse travels through the Bundle of His across the insulating fibrous skeleton of the heart and divides into right and left bundle branches, which distribute the signal to Purkinje fibers and ventricular myocardium. This pathway synchronizes ventricular contraction and supports efficient blood pumping; disruptions can cause atrioventricular or bundle branch block, making it important in cardiac physiology, electrocardiography, and clinical diagnosis.

Multiplexed Fluorescent Microarray for Human Salivary Protein Analysis Using Polymer Microspheres and Fiber-optic Bundles

We describe a procedure for profiling salivary proteins using multiplexed microsphere-based antibody arrays. Monoclonal antibodies were covalently linked to fluorescent dye-encoded 4.5 μm polymer microspheres using carbodiimide chemistry. The modified microspheres were deposited in fiber-optic microwells to measure protein levels in saliva using fluorescence sandwich immunoassays.

Here, we present a protocol for reconstituting microtubule bundles in vitro and directly quantifying the forces exerted within them using simultaneous optical trapping and total internal reflection fluorescence microscopy. This assay allows for nanoscale-level measurement of the forces and displacements generated by protein ensembles within active microtubule networks.
