Bundle Sheath Cells

Bundle sheath cells are specialized plant cells that encircle vascular bundles and help regulate exchange between leaf tissues and the plant’s transport system. In C4 plants, they form Kranz anatomy and receive four-carbon compounds from neighboring mesophyll cells; decarboxylation releases concentrated carbon dioxide, which Rubisco fixes in the Calvin cycle while limiting photorespiration. Their coordinated activity with mesophyll cells enables efficient carbon assimilation under high light, heat, and limited water availability. Studying bundle sheath cells clarifies how plants partition photosynthetic functions and supports research into crop improvement, stress tolerance, and engineering more efficient carbon-concentrating mechanisms.

We describe a procedure for profiling salivary proteins using multiplexed microsphere-based antibody arrays. Monoclonal antibodies were covalently linked to fluorescent dye-encoded 4.5 μm polymer microspheres using carbodiimide chemistry. The modified microspheres were deposited in fiber-optic microwells to measure protein levels in saliva using fluorescence sandwich immunoassays.

This manuscript details an optimized inoculation protocol that uses detached maize leaf sheaths for reproducible cytological, physiological, and molecular studies of maize interactions with fungal plant pathogens. The leaf sheaths facilitate real-time observation of cellular interactions between the living plant and fungus in unfixed tissues.

Optic Nerve Sheath Point of Care Ultrasound: Image Acquisition

Point-of-care ultrasound (POCUS) of the optic nerve sheath diameter (ONSD) has been shown to be useful in identifying patients with increased intracranial pressure (ICP). However, the non-standardized technique for this POCUS has hampered its use. We present a standardized image acquisition protocol for use in the acute care setting.
