Calcium Influx Imaging

Calcium influx imaging is a biological technique that visualizes changes in intracellular calcium concentration to monitor cell signaling and activity. It uses fluorescent calcium indicators, which change their optical properties when they bind Ca²⁺, allowing microscopy to track calcium entering cells through membrane channels or being released from internal stores. Researchers apply this approach to study neuronal communication, muscle contraction, secretion, and other calcium-dependent processes in living cells and tissues. By revealing when and where calcium signals occur, the method connects ion dynamics with cellular behavior and helps characterize signaling pathways, disease mechanisms, and responses to experimental treatments.

2025

This video describes a confocal microscopy imaging-based assay to measure mitochondrial calcium uptake in cultured cells. The assay uses the calcium-sensitive fluorescent dye Rhod-2/AM to measure the mitochondrial calcium in permeabilized cultured cells.

Assessing T-cell Receptor-Induced Calcium Influx Using a Calcium Indicator Dye

2025

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

(2020) This video demonstrates the real-time visualization of calcium influx in ventral midbrain neurons derived from mouse embryos. Using a viral vector to express a calcium indicator, the neurons are imaged under a confocal microscope to track changes in fluorescence intensity during spontaneous and neurotransmitter-induced calcium activity.

Calcium Imaging in Neurons

Calcium ions play an integral role in neuron function: They act as intracellular signals that can elicit responses such as altered gene expression and neurotransmitter release from synaptic vesicles. Within the cell, calcium concentration is highly dynamic due to the presence of pumps that selectively transport these ions in response to a variety of signals. Calcium imaging takes advantage of intracellular calcium flux to directly visualize calcium signaling in living neurons. This video begins...
