Cell Adhesion

Cell adhesion is the process by which cells attach to one another or to the extracellular matrix, providing the physical organization required for tissues to form and function. Specialized adhesion molecules, including cadherins and integrins, bind neighboring cells or matrix proteins and connect through intracellular proteins to the actin cytoskeleton; calcium ions help stabilize many cadherin-based contacts. These interactions regulate tissue architecture, cell signaling, migration, and mechanical strength during development and tissue repair. Studying cell adhesion helps explain how tissues maintain integrity and how disruptions can contribute to impaired development, inflammation, and disease progression.

Here, we present a protocol to continuously quantify cell adhesion and de-adhesion processes with high temporal resolution in a non-invasive manner by cell-substrate impedance and live cell imaging analyses. These approaches reveal the dynamics of cell adhesion/de-adhesion processes triggered by matrix modification and their temporal relationship to adhesion-dependent signaling events.
