Cord Blood Banking

Cord blood banking is the collection, processing, and cryogenic storage of blood left in the umbilical cord and placenta after birth, preserving a source of blood-forming stem and progenitor cells for future use. After delivery, trained personnel collect the blood without altering newborn care, then test it, separate or concentrate the cellular fraction, and freeze it under controlled conditions to maintain viability. Stored units may support hematopoietic stem cell transplantation for selected blood cancers, inherited blood disorders, and immune deficiencies, while also providing material for research on cell therapies and genetic disease. Its value depends on cell dose, sample quality, and donor-recipient compatibility.

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.
