Cryo Soft X-ray Tomography

Cryo soft X-ray tomography is a three-dimensional imaging method that visualizes intact, frozen-hydrated cells at nanometer-scale resolution, linking molecular organization with whole-cell architecture. In this technique, vitrified specimens are rotated in a soft X-ray beam, and transmitted X-ray projections are computationally reconstructed into a volume; differences in X-ray absorption provide natural contrast among cellular structures without sectioning or staining. The method enables researchers to map organelles, membranes, inclusions, and pathogen-host interactions in near-native cellular states. By combining structural detail with broad cellular context, cryo soft X-ray tomography supports studies of cell biology, infection, development, and disease mechanisms.

Tree Core Analysis with X-ray Computed Tomography

Here we show how to process tree cores with an X-ray computed tomography toolchain. Except for chemical extraction for some purposes, no further physical lab treatment is needed. The toolchain can be used for biomass estimations, for obtaining MXD/tree-ring width data as well as for obtaining quantitative wood anatomy data.

We present a protocol on how to utilize high-throughput cryo-electron tomography to determine high resolution in situ structures of molecular machines. The protocol permits large amounts of data to be processed, avoids common bottlenecks and reduces resource downtime, allowing the user to focus on important biological questions.
