Embryonic Day 8.5

Embryonic Day 8.5 (E8.5) is a defined developmental time point, typically referring to mouse embryos approximately 8.5 days after fertilization, when rapid tissue patterning and early organ formation occur. During this stage, gastrulation has established the body plan, while neural tube development, heart morphogenesis, axial patterning, and somite formation proceed through coordinated cell migration, differentiation, and signaling. Researchers use E8.5 embryos to examine gene function, embryonic morphogenesis, and the effects of genetic mutations or environmental exposures on development. Because many foundational structures form during this narrow window, E8.5 provides a valuable model for studying normal development and the origins of congenital abnormalities.

A System for ex vivo Culturing of Embryonic Pancreas

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

Embryonic stem (ES) cells are undifferentiated pluripotent cells, meaning they can produce any cell type in the body. This gives them tremendous potential in science and medicine since they can generate specific cell types for use in research or to replace body cells lost due to damage or disease. Origin ES cells are present in the inner cell mass of an embryo at the blastocyst stage, which occurs at about 3–5 days after fertilization in humans before the embryo is implanted in the uterus.

This video shows a procedure for isolating intact protoplasts from tissues of 14-day-old seedlings of Arabidopsis. Given that the isolated protoplasts remain intact for at least 96h and are isolated from seedlings instead of one-month-old mature plants, this procedure expedites assays requiring intact protoplasts.
