Endothelial Cell Migration

Endothelial cell migration is the directed movement of endothelial cells, which form the inner lining of blood vessels, to build, repair, and remodel the vascular network. In response to chemical and mechanical cues, cells polarize, reorganize their actin cytoskeleton, extend lamellipodia, attach to the extracellular matrix, and move through coordinated cycles of adhesion and contraction. This process is central to angiogenesis, wound healing, and vascular development, while abnormal migration can contribute to tumor growth, inflammation, and vascular disease. Studying endothelial cell migration helps researchers understand blood vessel formation and evaluate therapies, biomaterials, and drug candidates that alter vascular remodeling.

Investigation of early endothelial cell (EEC) migration is important to understand the pathophysiology of certain illnesses and to potentially identify novel strategies for therapeutic intervention. The following protocol describes techniques to assess cell migration that have been adapted for the investigation of EEC.

Primary cell culture is a useful technique for analyzing specific populations of cells, particularly from transgenic mouse embryos at specific developmental stages. Herein, embryonic ventricles are dissected and dissociated, and antibody-conjugated beads recognize and separate out the endothelial cells for further analysis.

Establishing a Co-Culture of Interneurons with Periventricular Endothelial Cells

This video demonstrates a technique for establishing a co-culture of interneurons with perivascular endothelial cells. The cells are added to a one-well insert placed inside a culture dish. After the cells adhere to the dish, the insert is removed, allowing the cells to proliferate and interact and form a co-culture.

An Introduction to Cell Motility and Migration

2023

Cell motility and migration play important roles in both normal biology and in disease. On one hand, migration allows cells to generate complex tissues and organs during development, but on the other hand, the same mechanisms are used by tumor cells to move and spread in a process known as cancer metastasis. One of the primary cellular machineries that make cell movement possible is an intracellular network of myosin and actin molecules, together known as “actomyosin”, which creates a...
