Enriched Fly Lysate

Enriched fly lysate is a concentrated biological extract prepared from fly tissues or embryos, providing cellular molecules for biochemical and molecular biology studies. Mechanical homogenization disrupts cells and releases proteins, nucleic acids, metabolites, and other intracellular components; clarification or fractionation can enrich selected material and remove larger debris. Researchers use these lysates to examine protein activity, gene expression, cellular signaling, and metabolic processes in Drosophila and related experimental systems. A PCR-based protocol was adapted to detect Cronobacter spp., Salmonella enterica, and Listeria monocytogenes from body surfaces and alimentary canals of individual wild-caught flies. The goal of this protocol is to detect and isolate bacterial pathogens from individual insects collected as part of an environmental sampling program during foodborne outbreak investigations.

Here we describe an isolation method that enhances the detection of subtle metabolic shifts in Drosophila melanogaster.
