Ex Vivo Vascular Reactivity

Ex vivo vascular reactivity is the study of how isolated blood vessels respond to physiological signals or vasoactive compounds outside the body, providing a controlled model of vascular function. In a tissue bath or organ-bath system, vessel segments are maintained under defined conditions and their diameter or force changes are measured as smooth muscle contracts or relaxes in response to agents that act on the endothelium or vascular muscle. This approach helps characterize vasoconstriction, vasodilation, endothelial function, and drug effects. It is widely used in biology and cardiovascular research to investigate vascular mechanisms, compare healthy and diseased vessels, and evaluate potential therapeutics.

Many vision-threatening ocular diseases are associated with dysfunctional retinal microvessels. Therefore, the measurement of retinal arteriole responses is important to investigate the underlying pathophysiological mechanisms. This article describes a detailed protocol for mouse retinal arteriole isolation and preparation to assess the effects of vasoactive substances on vascular diameter.

This is a guideline for constructing in vivo vascularized tissue using a microsurgical arteriovenous loop or a flow-through pedicle configuration inside a tissue engineering chamber. The vascularized tissues generated can be employed for organ regeneration and replacement of tissue defects, as well as for drug testing and disease modeling.
