Flow Cytometry

Flow cytometry is a laboratory method that rapidly measures physical and molecular characteristics of individual cells or particles suspended in fluid. As each particle passes in single file through one or more laser beams, detectors record light scattered by the cell and fluorescence emitted by labeled antibodies or other probes, allowing multiple features to be analyzed simultaneously. In biology, researchers use flow cytometry to identify cell populations, assess viability, quantify protein expression, and examine cell-cycle or immune responses. Fluorescence-activated cell sorting can further separate selected populations for culture, genetic analysis, or downstream experiments, making the technique valuable in basic research, disease studies, and biotechnology.

An efficient method to obtain highly purified viable meiotic fractions from mouse testis is described, which combines a refined cell dissociation protocol with fluorescent activated cell sorting (FACS). This method takes advantage of differences in the DNA content and nuclear density of discrete meiotic fractions.

The micronucleus (MN) assay is a well-established test for quantifying DNA damage. However, scoring the assay using conventional techniques such as manual microscopy or feature-based image analysis is laborious and challenging. This paper describes the methodology to develop an artificial intelligence model to score the MN assay using imaging flow cytometry data.

This article describes spectral cytometry, a new approach in flow cytometry that uses the shapes of emission spectra to distinguish fluorochromes. An algorithm replaces compensations and can treat auto-fluorescence as an independent parameter. This new approach allows for the proper analysis of cells isolated from solid organs.
