Fluorescent Dye Testing

Fluorescent dye testing is a laboratory technique that uses light-responsive molecules to detect, label, or measure biological structures and processes. When a dye absorbs light at a specific excitation wavelength, it emits light at a longer wavelength, allowing researchers to visualize or quantify the labeled target with fluorescence microscopy or other optical instruments. In biology, these dyes can reveal cell boundaries, organelles, nucleic acids, proteins, and changes in cellular conditions, supporting studies of cell function, localization, viability, and molecular interactions. The method provides sensitive, relatively rapid measurements for basic research, diagnostics, and evaluation of biological responses.

We described structural features of the Glia-neuromuscular synapses in a novel Inside-out tissue preparation of live fly larvae using fluorescent dyes with confocal microscopy. We labeled live neuron terminals with fluorescent primary antibodies to HRP, and also visualized the perisynaptic space with fluorescent Dextrans.
