Fluorescent Oligonucleotide Probes

Fluorescent oligonucleotide probes are short, synthetic nucleic acid sequences labeled with fluorescent molecules to detect specific DNA or RNA targets, making otherwise hidden molecular events visible in biological samples. A probe binds its complementary sequence through base pairing, and fluorescence from the attached reporter provides a signal that can be measured or imaged when the target is present; probe design and hybridization conditions help determine specificity. In biology, these probes support nucleic acid detection, gene-expression analysis, chromosome or cellular localization, and visualization of pathogen or genetic sequences, linking molecular identity to spatial or quantitative information in research and diagnostic applications.

We describe a multiplex method for the detection of microorganisms within a sample using oligonucleotide-coupled fluorescent beads. Amplicon from all organisms within a sample is hybridized to a panel of probe-coupled beads. A Luminex or Bio-Plex instrument is used to query each bead for bead type and hybridization signal.

We present a new fluorescence technique for selective in situ labeling of active phagocytic cells, which clear off cell corpses in stroke. The approach is important for assessing brain reaction to ischemia because only a small proportion of phagocytes present in ischemic brain participate in clearance of cell death.
