Frozen Section Embedding

Frozen section embedding is a rapid tissue-preparation technique that supports microscopic examination of biological specimens when results are needed faster than routine paraffin processing. In this method, fresh tissue is oriented in a mold, surrounded by a water-soluble embedding medium such as optimal cutting temperature compound, rapidly frozen, and sectioned in a cryostat to produce thin slices for staining. The approach preserves some cellular structures, lipids, enzymes, and molecular targets, making it useful for intraoperative assessment, histology, immunofluorescence, and research, although freezing artifacts and lower section quality can limit interpretation.

In this video, we demonstrate the sectioning of a frozen mouse brain tumor tissue using a cryostat. The frozen brain tissue sections so obtained are stored at low temperatures until further analysis.

Using Unfixed, Frozen Tissues to Study Natural Mucin Distribution

Unfixed frozen tissue samples embedded in Optimal Cutting Temperature medium (OCT) can be used to study natural distribution and glycosylation of secreted mucus. In this approach tissue processing is minimal and the natural presentation of glycolipids, mucins and glycan-epitopes is preserved. Tissue sections can be analyzed by immunohistochemistry using fluorescence or chromogenic detection.

Plastic sections maintain true tissue morphology in thin sections of tissue that can be immunostained with fluorescent secondary antibodies, making this method more useful than paraffin-embedded or frozen sections for many types of tissue. The method for staining, plastic embedding, and sectioning is demonstrated in this video.
