Frozen Sections

Frozen sections are thin slices of biological tissue prepared from rapidly frozen specimens for microscopic examination, allowing researchers and clinicians to evaluate cellular structure without lengthy fixation and processing. A cryostat maintains the tissue at a low temperature, enabling a specialized blade to cut sections that can be mounted on slides and stained, often soon after collection. In biology, this technique supports rapid assessment of tissue morphology, enzyme activity, lipid distribution, and antigen localization while preserving molecules that may be altered by chemical fixation. Frozen sections are especially valuable in pathology, developmental research, and studies requiring timely analysis of fresh or fragile biological samples.

The indirect immunofluorescence protocol described in this article allows the detection and the localization of proteins in the mouse mammary gland. A complete method is given to prepare mammary gland samples, to perform immunohistochemistry, to image the tissue sections by fluorescence microscopy, and to reconstruct images.
