Gfp Purification

GFP purification is a biochemical method for isolating green fluorescent protein from a mixture of cellular proteins, producing material suitable for analysis and experimentation. The process typically begins with cell lysis and clarification, followed by chromatography that separates GFP according to properties such as affinity, charge, or size; when GFP carries a polyhistidine tag, immobilized metal affinity chromatography binds the tag to nickel or cobalt, while washing removes contaminants and imidazole elutes the protein. Purified GFP supports studies of protein expression, folding, fluorescence, and stability, and provides a reliable reporter for investigating gene regulation, cellular localization, and molecular interactions in biology.

To purify zygotes of S. cerevisiae, haploid cells of opposite mating type were engineered to express red or green fluorescent proteins, co-incubated to allow zygote formation, and fractionated using a flow cytometry-based protocol. The highly-enriched fraction enables subsequent "-omic" studies, recovery of initial progeny, and systematic investigation of zygote morphogenesis.
