Immunoblotting Validation

Immunoblotting validation is the process of confirming that an immunoblot reliably detects the intended protein with appropriate specificity and sensitivity. In a typical workflow, proteins are separated by size using gel electrophoresis, transferred to a membrane, and probed with primary and labeled secondary antibodies; validation compares the observed band’s molecular weight, signal strength, and behavior in suitable controls. This approach helps identify nonspecific binding, antibody cross-reactivity, and technical variation. In biology, validated immunoblots support studies of protein expression, processing, localization, and changes associated with development, disease, or experimental treatment, strengthening the reproducibility and interpretation of molecular data.

Immunoblotting (western blotting) is a rapid and sensitive assay for the detection and characterization of proteins that works by exploiting the specificity inherent in antigen-antibody recognition. This video provides protocols for protein separation, blotting proteins onto membranes, immunoprobing, and visualization using chromogenic or chemiluminescent substrates.

We describe the use of quantitative immunoblotting to validate immunofluorescence histology coupled with image analysis as a means of quantifying a protein of interest in formalin-fixed, paraffin-embedded (FFPE) tissue samples. Our results demonstrate the utility of immunofluorescence histology for ascertaining the relative quantity of biomarker proteins in routine biopsy samples.

Reliability and Validity

2020

Reliability and validity are two important considerations that must be made with any type of data collection. Reliability refers to the ability to consistently produce a given result. In the context of psychological research, this would mean that any instruments or tools used to collect data do so in consistent, reproducible ways. Unfortunately, being consistent in measurement does not necessarily mean that you have measured something correctly. To illustrate this concept, consider a kitchen...
