In Vitro Assay

An in vitro assay is a laboratory test performed outside a living organism to measure a biological activity, interaction, or response under controlled conditions. The method isolates a biochemical reaction, cell system, or tissue sample and exposes it to defined substances or conditions, then quantifies an endpoint such as enzyme activity, cell viability, binding, or gene expression against appropriate controls. In biology, in vitro assays support studies of molecular mechanisms, disease processes, drug efficacy, and toxicity while reducing experimental complexity and enabling precise comparisons. Results can guide further cellular, animal, or clinical research, although they may not fully reproduce conditions in a living system.

The second method is the transwell migration and invasion assay that assesses the chemotactic and invasive capacity of cells.

Unmodified and hyperphosphorylated tau proteins were used in two in vitro aggregation assays to reveal the hyperphosphorylation-dependent fast aggregation kinetics. These assays pave the way for future screens for compounds that can modulate the propensity of hyperphosphorylated tau to form fibrils that underlie the progression of Alzheimer’s disease.
