Label-free Volumetric Imaging

Label-free volumetric imaging is a set of microscopy methods that generates three-dimensional views of biological specimens without fluorescent or chemical labels, preserving native structure and reducing sample preparation. These techniques detect intrinsic optical properties, such as variations in refractive index, light scattering, absorption, or phase, and computationally reconstruct them across successive planes or throughout a specimen. In biology, label-free volumetric imaging supports noninvasive observation of living cells, tissues, and developing organisms over time. It can reveal morphology, dynamics, and cell interactions while minimizing phototoxicity and perturbation, making it valuable for developmental biology, disease research, and quantitative cell analysis.

This protocol presents a step-by-step guide for the Simultaneous Label-free Autofluorescence Multi-harmonic (SLAM) microscopic technique, including details on how to generate the laser light source, prepare a tissue sample, conduct imaging, and analyze the data. SLAM advances nonlinear microscopy by measuring four complementary label-free contrasts to investigate the tissue microenvironment.
