Linker Histone H1

Linker histone H1 is a chromatin-associated protein that binds DNA between nucleosomes and helps organize the genome within the nucleus. Its central globular domain recognizes the nucleosome while flexible, positively charged regions contact linker DNA, stabilizing DNA entry and exit sites and promoting tighter chromatin folding; phosphorylation can alter these interactions and chromatin accessibility. Through this mechanism, histone H1 influences whether regulatory proteins and transcription machinery can reach specific genes. Studying H1 supports research on gene regulation, genome organization, cell differentiation, and development, while changes in its abundance or modification state can provide insight into abnormal chromatin structure and disease.

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.
