Luciferase Inhibitors

Luciferase inhibitors are compounds that reduce the light-producing activity of luciferase enzymes, making them important tools for studying bioluminescence and validating reporter-based experiments. Depending on the enzyme and compound, inhibition can occur when a molecule binds the luciferase active site, competes with luciferin or ATP, disrupts catalytic oxidation, or quenches the emitted light. In biology, these inhibitors help characterize luciferase structure and kinetics, identify assay interference, and control signal strength in gene-expression and drug-screening studies. Understanding their selectivity also improves interpretation of bioluminescent assays and supports the design of more reliable imaging and biosensor systems.

Here, we present a method for screening anti-hepatitis B viral agents that inhibit the HBx-DDB1 interaction using a split luciferase assay system. This system allows easy detection of protein-protein interactions and is suitable for identifying inhibitors of such interactions.
