Liver Endothelial Cells

Liver endothelial cells are specialized cells that line the blood vessels of the liver, especially the hepatic sinusoids, where they regulate exchange between circulating blood and liver tissue. Unlike many vascular endothelial cells, liver sinusoidal endothelial cells contain fenestrations, or small pores, and typically lack a continuous basement membrane; this arrangement facilitates the passage of plasma solutes while helping control hepatocyte exposure to blood-borne signals and substances. Their structure and function are central to hepatic filtration, metabolism, inflammation, and disease research, including studies of liver injury, fibrosis, drug delivery, and engineered liver models.

The study of liver sinusoidal endothelial cells (SECs) must be performed with primary cells obtained from the animal as no cell lines exist. This method relies on liver digestion and differential centrifugation for SEC purification for subsequent culturing and experimentation.

The fenestrated liver sinusoidal endothelial cell is a biologically important filter system that is highly influenced by various diseases, toxins, and physiological states. These changes significantly impact on liver function. We describe methods for the standardisation of the measurement of the size and number of fenestrations in these cells.

Isolation and Characterization of Mouse Primary Liver Sinusoidal Endothelial Cells

Here we outline and demonstrate a protocol for primary mouse liver sinusoidal endothelial cell (LSEC) isolation. The protocol is based on liver collagenase perfusion, nonparenchymal cell purification by low-speed centrifugation, and CD146 magnetic bead selection. We also phenotype and characterize these isolated LSECs using flow cytometry and scanning electron microscopy.

Purification of Hepatocytes and Sinusoidal Endothelial Cells from Mouse Liver Perfusion

The goal of this protocol is to obtain high viability and high yield of hepatocytes and sinusoidal endothelial cells from liver. This is accomplished by perfusing the liver with a type IV collagenase solution via the portal vein, followed by differential centrifugation to obtain hepatocytes and sinusoidal endothelial cells.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
