Murine Fibroblast Cells

Murine fibroblast cells are connective-tissue cells isolated from mice that produce and maintain extracellular matrix, making them useful models for studying tissue structure and repair. They synthesize components such as collagen and other matrix proteins, while responding to signals from growth factors, cytokines, and nearby cells that regulate proliferation, migration, and differentiation. In biology, cultured murine fibroblasts support research on wound healing, fibrosis, inflammation, cell signaling, and genetic function. Their defined species origin and adaptability to laboratory culture also make them valuable for investigating cellular mechanisms and evaluating how mutations, environmental conditions, or experimental treatments alter connective-tissue behavior.

Here we present a protocol for the direct conversion of murine embryonic fibroblasts into fully functional and stable trophoblast stem cells by ten day over-expression of Tfap2c, Gata3, Eomes and Ets2.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.

Experimental Generation of Carcinoma-Associated Fibroblasts (CAFs) from Human Mammary Fibroblasts

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Generating Neurons by Reprogramming Human Dermal Fibroblast Cells

2025

The video demonstrates the generation of neurons from adult human dermal fibroblasts (aHDFs). The aHDFs are reprogrammed using a lentiviral vector carrying genes for neuronal transcription factors and short hairpin RNAs (shRNAs). The shRNAs inhibit the expression of a neuronal transcriptional repressor, and the neuronal transcription factors activate genes to differentiate aHDFs into neurons. These reprogrammed aHDFs are cultured in an early and a late neuronal conversion medium to generate...
