Muscle Strip Preparation

Muscle strip preparation is an ex vivo technique that isolates a defined section of muscle for controlled study of its structure and contractile function. The strip is carefully dissected to preserve fiber orientation, mounted in a tissue bath containing oxygenated physiological solution, and connected to a force transducer; electrical or chemical stimulation then elicits contraction while force is recorded. Researchers can vary stimulation frequency, muscle length, or pharmacological conditions to assess contractility, fatigue, and responses to signaling molecules. In biology, this preparation provides a direct model for investigating muscle physiology, neuromuscular function, disease-related changes, and the effects of potential therapeutics.

Development of a Colloidal Gold-based Immunochromatographic Test Strip for Detection of Cetacean Myoglobin

This protocol describes the development of two IgG class monoclonal antibodies (mAbs) strongly reactive to myoglobin of cetaceans. These mAbs are applied on a colloidal gold immunochromatographic test strip based on the sandwich format to differentiate the Mb of cetaceans from seal and other animals.

The Mouse Cremaster Muscle Preparation for Intravital Imaging of the Microcirculation

A tissue preparation is described for visualization and experimental manipulation of the living microcirculation. In anesthetized male mice, the thin, highly vascularized cremaster muscle is prepared for intravital microscopy to study microvascular networks including arterioles, capillaries and venules. This preparation is readily adapted for rats and hamsters.
