Neutral Lipids

Neutral lipids are hydrophobic molecules that lack a substantial net charge and serve as major forms of energy storage in biological systems. In cells, they commonly include triacylglycerols and cholesteryl esters, formed when fatty acids become esterified to glycerol or cholesterol, reducing their polarity and promoting packaging into lipid droplets. Enzymes such as lipases hydrolyze these ester bonds when energy is needed, releasing fatty acids for oxidation or other metabolic pathways. Studying neutral lipids helps explain fuel storage, membrane lipid balance, obesity, metabolic disease, and lipid droplet biology, while fluorescence imaging and mass spectrometry reveal their abundance, localization, and turnover.

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
