New Culture Method

New culture methods in biology are laboratory approaches for growing and maintaining cells, tissues, or microorganisms under controlled conditions, enabling researchers to study living systems outside their original environment. Typically, a sample is introduced into a suitable nutrient medium, then supplied with carefully regulated temperature, pH, oxygen, and sterile conditions so target organisms can multiply while contamination is limited. These methods support investigations of physiology, development, disease, and microbial behavior, while improving the efficiency and reproducibility of laboratory research. Their refinement can also expand opportunities for screening treatments, producing biological materials, and modeling complex biological processes.

This video demonstrates New culture, a method by which chick embryos are cultured outside the egg for up to 24 hr. This method enables one to study early development (primitive streak to 14 som.), a period corresponding to E7-9 in mouse. Applications of this technique include electroporation, in situ hybridization and immunohistochemistry.

The Method of Rodent Whole Embryo Culture using the Rotator-type Bottle Culture System

Whole embryo culture technique allows us to culture mouse and rat embryos ex vivo condition during limited periods corresponding to midgestation stages. In this video protocol, we demonstrate our standard procedures of rat whole embryo culture after E12.5 using the rotator-type bottle culture system.

Cell Culture Methods for New Approach Methodologies

2026

Mammalian and other animal-derived cell culture systems are widely used in biomedical research as New Approach Methodologies (NAMs) that reduce reliance on animal testing while enabling controlled investigation of cellular responses. These in vitro systems provide reproducible platforms for studying disease mechanisms, drug activity, toxicity, and neuronal function under defined laboratory conditions. Establishment and Maintenance of Cell Cultures To establish a culture, cells are seeded into...

A Method for Culturing Embryonic C. elegans Cells

We describe here a revised protocol for large-scale culture of embryonic C. elegans cells. Embryonic C. elegans cells cultured in vitro using this method, appear to differentiate and recapitulate the expression of genes in a cell specific manner. Techniques that require direct access to the cells or isolation of specific cell types from the other tissues can be applied on C. elegans cultured cells.

A Novel Ex vivo Culture Method for the Embryonic Mouse Heart

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.
