Paraffin Block Preparation

Paraffin block preparation is a histology technique that preserves biological tissue by embedding it in solid paraffin wax, creating a stable block for thin-section microscopy. The process typically involves fixation to limit degradation, dehydration to remove water, clearing to replace the dehydrating agent, and infiltration with molten paraffin; as the wax cools, it hardens around the specimen and supports sectioning with a microtome. Researchers can then stain tissue sections to examine cellular organization, morphology, and disease-related changes, making paraffin blocks useful for teaching, routine pathology, biological research, and long-term sample archiving.
