Protein Interaction Sites

Protein interaction sites are specific regions on proteins where they bind other proteins or biomolecules, enabling cellular communication, organization, and regulation. These sites form through complementary molecular features, including shape, charge, hydrophobicity, hydrogen bonding, and short recognition motifs; binding may also involve conformational changes that create a functional interface. Identifying interaction sites helps explain protein complexes, signaling pathways, enzyme regulation, and the effects of disease-associated mutations. In biology and drug discovery, researchers use structural analysis, mutagenesis, biochemical assays, and computational modeling to map these regions and design molecules that strengthen, block, or modify protein interactions.

Identifying Protein-protein Interaction Sites Using Peptide Arrays

Peptide array screening is a high throughput assay for identifying protein-protein interaction sites. This allows mapping multiple interactions of a target protein and can serve as a method for identifying sites for inhibitors that target a protein. Here we describe a protocol for screening and analyzing peptide arrays.
