Retinal Explants

Retinal explants are pieces of retinal tissue maintained outside the organism under controlled laboratory conditions, preserving interactions among neurons, glial cells, and supporting layers. Researchers typically dissect the retina, place it on a culture support or in specialized medium, and provide nutrients and environmental conditions that sustain tissue organization while enabling direct observation and experimental manipulation. This ex vivo model supports studies of retinal development, neuronal survival, degeneration, injury, and responses to therapeutic compounds or genetic interventions. Because explants retain aspects of native retinal structure without the complexity of a whole animal, they help connect cellular mechanisms with tissue-level outcomes in biology and vision research.

Whole Retinal Explants from Chicken Embryos for Electroporation and Chemical Reagent Treatments

This protocol describes a method to dissect, experimentally manipulate and culture whole retinal explants from chicken embryos. The explant cultures are useful when high success rate, efficacy and reproducibility are needed to test the effects of plasmids for electroporation and/or reagent substances, i.e., enzymatic inhibitors.

Zebrafish retinal regeneration has mostly been studied using fixed retinas. However, dynamic processes such as interkinetic nuclear migration occur during the regenerative response and require live-cell imaging to investigate the underlying mechanisms. Here, we describe culture and imaging conditions to monitor Interkinetic Nuclear Migration (INM) in real-time using multiphoton microscopy.

Organotypic Retinal Explant Cultures from Macaque Monkey

Retinal explants obtained from wild-type macaques were cultured in vitro. Retinal degeneration and the cGMP-PKG signaling pathway was induced using the PDE6 inhibitor zaprinast. cGMP accumulation in the explants at different zaprinast concentrations was verified using immunofluorescence.
