Ruthenium Red

Ruthenium red is a polycationic inorganic dye used in biology to label acidic, negatively charged structures and to probe ion transport. Its positively charged ruthenium complex binds anionic groups in materials such as pectins, mucopolysaccharides, and other extracellular or membrane-associated components, while also inhibiting several calcium-permeable ion channels under experimental conditions. These properties make ruthenium red useful for electron and light microscopy, including studies of cell walls, extracellular matrices, and organelle ultrastructure. Researchers also apply it as a pharmacological tool to investigate calcium signaling, mechanosensation, and channel function, although its broad activity requires careful interpretation and appropriate controls.

Ruthenium phosphine complexes are widely used for homogeneous catalytic reactions such as hydrogenations. The synthesis of a series of novel tridentate ruthenium complexes bearing the N-triphos ligand N(CH2PPh2)3 is reported. Additionally, the stoichiometric reaction of a dihydride Ru–N-triphos complex with levulinic acid is described.

Here we present techniques to measure red cell deformability and cellular heterogeneity by ektacytometry. These techniques are applicable to general investigations of red cell deformability and specific investigations of blood diseases characterized by the presence of both rigid and deformable red cells in circulation, such as sickle cell anemia.
