Slos Spheres

SLOS spheres are three-dimensional cellular models used to study Smith-Lemli-Opitz syndrome, a developmental disorder caused by impaired cholesterol biosynthesis. They are formed by aggregating cells with deficient or disrupted DHCR7 activity, which limits the conversion of 7-dehydrocholesterol to cholesterol and produces characteristic sterol imbalances within the sphere. By recreating cell interactions and metabolic conditions in a compact tissue-like structure, SLOS spheres help researchers examine how altered cholesterol homeostasis affects development, signaling, and cell function. These models can support disease-mechanism studies, comparison of patient-derived and control cells, and evaluation of potential therapeutic strategies.
