Stromal Tissue Removal

Stromal tissue removal is a biological sample-preparation process that separates supportive connective tissue from the functional cells or structures it surrounds, enabling more focused analysis of tissue composition and behavior. Researchers typically combine careful dissection with mechanical dissociation or enzymatic digestion, using enzymes such as collagenase to break down extracellular-matrix components while limiting damage to the target cells. This approach supports primary cell isolation, histological assessment, organoid preparation, and studies of interactions between stromal and parenchymal or epithelial compartments. By reducing tissue complexity, stromal tissue removal can improve cell characterization and clarify how the microenvironment influences development, repair, and disease.

In this video, we demonstrate the processing of FFPE or formalin-fixed paraffin-embedded sections obtained from gastrointestinal tumor tissue. The deparaffinization and rehydration steps allow the processing of the tissue specimen for subsequent analytical applications.
