Taxonomic Resolution

Taxonomic resolution is the level of precision at which an organism or biological sample can be identified and classified, from a broad group such as a family to a specific species or strain. It depends on the distinguishing information available, including morphological traits, genetic markers, DNA sequences, and the quality of taxonomic reference data used for comparison. Higher resolution can reveal closely related species, cryptic diversity, and shifts in community composition that broader classifications may obscure. In biology, taxonomic resolution supports biodiversity surveys, ecological monitoring, conservation planning, and the interpretation of environmental samples, while also helping researchers assess the limits of their identification methods.

2025

The protocol presented here aims to guide the users on the computer-based processing of cytochrome c oxidase subunit I (COI) gene sequences generated from beetles, such that the species clustering hypothesis called molecular operational taxonomic units (MOTUs) can be generated from DNA.

Optical-resolution photoacoustic microscopy (OR-PAM) is an emerging technology capable of imaging optical absorption contrasts in vivo with cellular resolution and sensitivity. Here, we provide a visualized instruction on the experimental protocols of OR-PAM, including system configuration, system alignment, typical in vivo experimental procedures, and functional imaging schemes.

Chromatographic Resolution

In chromatography, a solute moves through a chromatographic column and tends to spread, forming a Gaussian-shaped band. The longer the solute spends in the column, the broader the band becomes. The broadening can lead to overlaps within the column, affecting separation effectiveness. The effectiveness of separation can be evaluated by determining the level of separation between two neighboring peaks in a chromatogram, which represents the individual components of a sample. In chromatography,...

We describe the preparation of three test samples and how they can be used to optimize and assess the performance of STORM microscopes. Using these examples we show how to acquire raw data and then process it to acquire super-resolution images in cells of approximately 30-50 nm resolution.

High Resolution Phonon-assisted Quasi-resonance Fluorescence Spectroscopy

The manuscript describes a method of phonon-assisted quasi-resonant fluorescence spectroscopy that incorporates both laser-limited resolution and photoluminescence (PL) spectroscopy. This method utilizes optical phonons to provide linewidth-limited resolution spectra of atom-like semiconductor structures in the energy domain. The method is also easily realized with a single spectrometer optical spectroscopy setup.
